Kombucha SCOBY Science: Probiotics, GABA Synthesis & Fermentation
Biochemical breakdown of Symbiotic Cultures of Bacteria and Yeast (SCOBY), organic acid conversion pathways, and their impact on the gut microbiome.
Kombucha is sweetened tea that has been eaten by a microbial community and turned into something sour. That is the whole of it, and it is genuinely interesting — but almost everything claimed for it beyond "fermented tea, tastes good, low in sugar" runs ahead of what has been shown.
What a SCOBY actually is
The floating disc is a cellulose pellicle — a biofilm produced by acetic acid bacteria as a by-product of their metabolism. It is not the culture. It is a structure the culture happens to build, and most of the microbial activity is in the liquid, not the disc.
The community inside is a partnership:
Yeasts — typically Saccharomyces, Brettanomyces, Zygosaccharomyces — hydrolyse sucrose into glucose and fructose, then ferment those into ethanol and carbon dioxide.
Acetic acid bacteria — chiefly Komagataeibacter (formerly Gluconacetobacter) — oxidise that ethanol into acetic acid, and glucose into gluconic acid.
The two are coupled. The yeasts produce the substrate the bacteria need; the bacteria keep ethanol low enough that the yeasts are not inhibited. It is a stable loop, which is why a healthy culture can run for years.

The claims worth being sceptical about
"It detoxifies your liver via glucuronic acid." This is the most repeated kombucha claim and the weakest. Two separate problems. First, analyses disagree on whether meaningful glucuronic acid is present at all — several find little or none. Second, and more fundamentally, glucuronidation is a liver process that runs on the body's own UDP-glucuronic acid, synthesised internally. Drinking glucuronic acid does not feed that pathway. The claim does not work even if the compound is there.
"It is a probiotic." The organisms in a SCOBY are acetic acid bacteria and fermentative yeasts. Clinical probiotic research has overwhelmingly studied Lactobacillus and Bifidobacterium strains at defined doses. Kombucha's organisms are not those, and no dose is defined. Calling it a probiotic stretches a regulated term past what has been tested.
"The GABA relaxes you." Some lactic acid bacteria produce GABA during fermentation and it has been detected in some kombucha. But GABA crosses the blood-brain barrier poorly, which is why oral GABA supplements have such thin evidence. A calming effect from a glass of kombucha is not an established finding.
Kapp and Sumner's systematic review of the human evidence reached a blunt conclusion: the claimed benefits rest largely on animal and in-vitro work, and the human trial base is close to absent.
What can be said fairly
It is a low-sugar fermented drink — most of the added sugar is consumed by the culture — with the polyphenol content of the tea it was made from. It is pleasant, it is interesting, and for people replacing soft drinks with it, that swap is a real improvement on its own terms.
The base tea matters more than most brewers treat it. A black tea base ferments differently from a green one, and the polyphenol profile of the finished drink follows the leaf.
The risks, which do not get mentioned enough
These are documented, not hypothetical.
- Over-fermentation produces a drink acidic enough to be unpleasant and, in extreme cases, to cause gastrointestinal upset.
- Lead leaching from glazed ceramic vessels has caused documented poisoning. Ferment in glass. This is the single most important safety rule in home brewing.
- Contamination by mould is a real failure mode. A fuzzy growth is not a healthy SCOBY.
- Alcohol content in home brews can exceed the 0.5% commercial threshold, which matters in pregnancy, in recovery, and for children.
- Immunocompromised drinkers should be cautious with any unpasteurised live-culture product.
Brewing it well
- Glass vessel. Never glazed ceramic, never metal for the primary ferment.
- Clean hands, clean equipment, breathable cover.
- Seven to fourteen days at room temperature for the primary ferment; taste from day five.
- Stop when the sourness is where you like it. There is no virtue in pushing further.
- Second ferment in sealed bottles for carbonation — and burp them, because pressure builds fast.
Frequently asked
Does kombucha have caffeine? Yes, carried over from the tea, though reduced by fermentation. Typically 10–25 mg per serving.
Is home-brewed safer or riskier than shop-bought? Riskier, in the specific senses above: variable alcohol, contamination risk, and vessel choice. Commercial production is pasteurised or tested.
Should I drink it on an empty stomach? No particular reason to, and its acidity makes it a poor choice for anyone prone to reflux.
The pH curve is the safety mechanism
This is the part of kombucha brewing that is genuinely important and gets the least attention.
Sweetened tea starts around pH 5, which is hospitable to a great many things you do not want. Within a few days of a healthy ferment it drops to roughly 3.0-3.5, and below about pH 4.2 most foodborne pathogens cannot establish. The acidity is not a by-product of the process; it is the reason the process is safe.
Which makes the starter liquid the single most important ingredient. Adding roughly 10% mature, sour kombucha to a fresh batch drops the pH immediately, so the vulnerable window is measured in hours rather than days. A batch started with a pellicle and no starter liquid sits at hospitable pH while the culture wakes up, and that is when contamination happens.
Cheap pH strips settle the question. If a batch has not dropped below 4.2 within two or three days, something is wrong: too cold, too little starter, or a culture that has been stored badly. Discard rather than persist.
How much sugar is actually left
"Kombucha is sugar-free" is wrong, and "kombucha is full of sugar" is also wrong.
A typical batch starts near 50-70 g of sugar per litre. The yeasts consume most of it over the primary ferment, and finished commercial kombucha commonly lands somewhere around 20-40 g/l, which is roughly 2-4 g per 100 ml. That is well under most soft drinks and well over water.
Two things move the number. A longer ferment leaves less sugar and more acid — the two trade against each other, so a bone-dry kombucha is also a sharp one. And the second ferment adds sugar back, because juice or fruit is what feeds the carbonation. A heavily flavoured bottle can end up close to where it started.
If you are tracking intake, read the label rather than the category.
Temperature, and what goes wrong at each end
The culture works best somewhere around 24-29°C, and it tells you clearly when it is unhappy.
Too cold, below about 20°C, and everything slows. The pH drop is sluggish, which extends the window in which mould can take hold. This is why winter batches fail more often than summer ones.
Too warm, above about 32°C, and the yeasts outrun the bacteria. You get a boozier, more solvent-smelling brew, more yeast strands, and a thin pellicle.
A brewing belt or simply a warmer shelf solves the first. The second usually solves itself by moving the vessel off a sunny windowsill.
Second fermentation, and bottle safety
Carbonation comes from a sealed second ferment: bottle the finished kombucha with a little sugar or fruit, leave it at room temperature for two to four days, then refrigerate to stop it.
The pressure this generates is not trivial, and this is where home brewing hurts people.
- Use bottles rated for pressure. Swing-top bottles designed for beer or a purpose-made kombucha bottle. Not a repurposed juice bottle, not decorative glass.
- Burp them daily. Open briefly to release pressure, especially in warm weather.
- Refrigerate to halt it. Cold does not stop fermentation entirely but slows it enough to be safe.
- Watch the alcohol. The second ferment is sealed, so ethanol accumulates rather than oxidising off. Home brews can exceed the 0.5% threshold that commercial products are held to.
If a bottle hisses aggressively or the cap bulges, chill it before opening, and open it in a sink.
Jun, and the neighbours
Jun is the closest relative: green tea and honey rather than black tea and sugar, with a culture adapted to that substrate. It ferments faster, at a lower temperature, and finishes lighter and more floral. It is sometimes marketed as "the champagne of kombucha", which is marketing, but the drink is genuinely distinct. Water kefir looks similar and is not related. It uses a different culture — translucent grains rather than a pellicle — ferments sugar water rather than tea, and contains no tea polyphenols at all.
Tepache, kvass and switchel all sit in the same part of the shelf and share the appeal: sour, lightly fermented, low in sugar relative to soft drinks. None of them has the clinical evidence base their labels imply either, and that is fine as long as nobody is claiming otherwise.
